DNAWorks v3.2.4 is an automated tool for designing oligonucleotides for PCR-based gene synthesis. Given a protein and/or nucleotide target sequence plus a codon-frequency table, DNAWorks performs Monte-Carlo optimisation that simultaneously balances annealing temperature, codon usage, repeat content, misprime potential, GC/AT content, and user-supplied pattern constraints, then partitions the optimised gene into a thermodynamically balanced set of overlapping oligonucleotides ready for PCR assembly.
